In the latest episode of the Alpha Human Podcast, Lawrence shares his experience of delving into the medical literature for six months after being told that other than surgery, there were no effective treatments for his type of cancer. The results of his genetic analysis showed that his tumor had a mutation of the DDX41 gene, responsible for creating a protein that suppresses certain oncogenes, such as KIT. What's more, he discovered that his tumor not only expressed higher levels of the kinase receptor protein C-KIT compared to normal cells, but he also learned has a KIT mutation. This is noteworthy because KIT activating mutations are typically never seen in chromophobe renal cell carcinoma (ChRCC), which is Lawrence's cancer type. This finding could have significant implications for targeted therapies, such as Tyrosine Kinase Inhibitors, which although usually ineffective in chRCC, may work in Lawrence's case due to his unique mutation profile.

The episode also highlights a scientific paper from 2019 that reported on a patient with metastatic ChRCC who had an exceptional response to Sunitinib, despite tyrosine kinase inhibitors (TKIs) not being typically effective for this type of cancer. The patient had increased expression of VEGF-C and VEGF-B, which could be utilized as biomarkers to guide ChRCC patients toward TKI therapy. Lawrence stresses the importance of doing your own research and taking control of your health, especially when dealing with serious medical conditions.

Decoding the Origin of Cancer

In this episode of the Alpha Human Podcast, Lawrence Rosenberg dives into the fascinating connection between iron overload and cancer. Iron is an essential micronutrient that plays a crucial role in various biological processes, including energy production, DNA synthesis, and immune system function. 

However, excess iron is also toxic and iron accumulation is typically seen in renal cell carcinoma (as well as other cancers), and increased iron metabolism is associated with the malignant transformation of cells, cancer progression, drug resistance, and immune evasion.  In fact, individuals working in iron and steel occupations have a higher risk of developing kidney cancer.

With that said, Lawrence also introduces a recently discovered form of programmed cell death called ferroptosis, which is induced by elevated intracellular iron levels and subsequent depletion of the antioxidant glutathione. Ferroptosis represents a natural mechanism to eliminate cancer cells, as well as those cells deficient in key nutrients or damaged by infection or ambient stress. While ferroptosis is beneficial in removing problematic cells, it should also be noted that dysregulation or excessive activation can contribute to disease pathogenesis, including neurodegenerative disorders.

Interestingly, Lawrence highlights the unique characteristic of chromophobe renal cancer cells in having strikingly elevated levels of glutathione, which acts as a reactive oxygen species scavenger that protects these cells from ferroptosis. And although this metabolic dependency on glutathione enables chromophobe renal cell carcinoma (as well as a number of other cancers) to survive despite iron overload and oxidative stress, it may also turn out to be cancer's Achilles heal.

Iron: The Double-Edged Sword in Cancer

In this episode of The Alpha Human Podcast, Lawrence delves into drugs that can induce Ferroptosis, a form of cell death characterized by the accumulation of lipid peroxides and iron-dependent reactive oxygen species, as this might be an Achilles' heel for cancers that exhibit iron overload.

In this episode of the Alpha Human Podcast, Lawrence Rosenberg discusses the potential link between the consumption of peanuts and cancer metastasis. Peanuts contain peanut agglutinin (PNA), a lectin that binds to a complex sugar known as the T-antigen, a glycan bound to proteins found on certain cancer cells. This interaction mimics the actions of galectin-3, an endogenously produced human lectin (often overexpressed in cancer patients) that can bind to dysregulated glycoproteins on the membranes of cancer cells leading to enhanced cellular adhesion, invasion, migration, metastasis, angiogenesis, and immune evasion.

Additionally, PNA has been shown to interact with endothelial cells and trigger the secretion of metastasis-promoting cytokines that can lead to increased adhesion between tumor cells and blood vessel walls, facilitating tumor cell metastasis.

His decorations include 2 Bronze Stars w/ Valor, the Purple Heart, the Defense Meritorious Service Medal, the Joint Commendation Medal, and the Navy Commendation Medal w/ Valor x4.
   
After his time with the SEALs,  Dan became an instructor at the United States Naval Academy where he developed leadership programs and trained Navy and Marine Corps officers. He earned a Master’s degree in Organizational Leadership from Georgetown University and is the Founder of 3LX, a leadership consultancy and coaching business. 

Dan Luna's journey to becoming a Navy SEAL began as an only child fascinated with The Hardy Boy mysteries. He romanticized the idea of joining a brotherhood and going through a rite of passage, and the more books he read the more the idea of testing himself and the intrigue of becoming involved in big, international affairs appealed to him. His search for a crucible to challenge himself led him to look for the hardest program possible to forge him into the man he was looking to become. Dan considered law enforcement and the military, but nothing seemed to compare to the hell one must go through to become a Navy SEAL, the program known as BUDs (Underwater Demolition/SEAL Training). Dan graduated BUDS class 228, the class featured in the book The Warrior Elite: Forging of SEAL Class 228 by Dan Couch -- BUDS 228 is also the same class that produced Marcu Lutrell, the lone survivor of Operation Red Wings.

In this episode of the Alpha Human Podcast, Dan talks about the struggle of transitioning from a 20-year career with the SEALs and military as a warfighter handling presidential-level missions, to the metronome-like tempo of civilian life. "What you do as an occupation, it goes into your being, and that being (becomes) your character, your personality, your actions, and how you move day-to-day, and how you look at the world, and how you perceive things. So to sort of move from that from (being) covert to overt for me ... I felt like a Martian. I felt like I was on another planet." 

Dan explains how his epiphany for how to navigate and succeed in the foreign terrain of civilian life came when he asked himself a different question from the one that had been vexing him ... "instead of thinking about doing and I had a lot of issues with that what I started asking was different question was, who do I want to be?"

We also talk about building a "Feeder Mindset" and avoiding falling into the trap of the "Receiver mentality," how this applies to success in all realms of life, a fascinating concept found within Sayoc Kali, a fighting system for using edged weapons (blades) in combat that Luna and many in the SpecOps/shooter community practice.

Ex-Navy SEAL Dan Luna: Leading Through Volatility

Tony Schwalm is the author of The Guerrilla Factory: The Making of Special Forces Officers – The Green Berets. Schwalm is uniquely qualified to tell the story of what it takes to forge unconventional warriors, as he helped redesign the very test of strength, stamina, and wits known as the Q Course, the Special Forces Qualifications program, which is the formal training program for entry into the United States Army Special Forces. Schwalm is a former lieutenant colonel with the U.S. Army Special Forces and his military career spans some 30 years. His journey has seen him transition from a tank commander in Operation Desert Storm (the first Iraq war) where he was awarded the Bronze Star for Valor, to becoming a Green Beret and later a commander of Special Forces officer training at Ft. Bragg. His assignments around the globe include leadership positions in combat operations, humanitarian missions, and counter-drug operations. After the attacks of September 11, 2001, Tony served with U.S. Special Operations Command creating state-of-the-art technology architectures to support Special Operations Forces. From there, as a civilian working with the Department of the Army, Tony led a team of social scientists in support of the Combined Joint Special Operations Task Force in Afghanistan, supporting counterinsurgency operations.

While much is known about the direct action-focused elements of the USA’s Special Operations Forces (SOF) such as the Navy Seals and Army Rangers, little is known about America’s unconventional warriors the Green Berets. The disparity in the attention given to these elite soldiers is no surprise given that much of the films, novels, and publicity generated around the exploits of America’s special ops is focused on the hyper-muscular actions of the special operators that fight the Global War on Terror (GWOT), the elite warriors responsible for short-duration strikes and other small-scale offensive actions, everything from eliminating high-value targets, raids and sabotage through to hostage rescue. And although the U.S. Army Special Forces (Green Berets) have direct action capabilities, and are certainly involved in such missions more than at any time in history since the war on terror began 20 years ago, their formation by President John F Kennedy to wage “unconventional war”,” as they did everywhere from Vietnam through to Panama, is where their deadliest and most effective skill set lies. Unconventional Warfare is rooted in the concept of insurgency, guerilla warfare, subversion, and the use of these strategies in support of a country’s resistance movement, a proxy army that allows the U.S. to topple a dictator, occupying power or a hostile government. And, there is no force better at fomenting a foreign insurgency than the Green Berets, their training and legacy are borne out of an uncanny, almost preternatural proficiency in the art killing the enemy from within.

Delta Force Operator and Green Beret Dale Comstock - A Modern Day Spartan

Priority number one was taking down Pablo Escobar, the populist, charismatic head of the Medellin Cartel who also led the terrorist group known as "Los Extraditables," an alliance of the country's drug lords whose goal was to pressure the Columbian government into ending its extradition treaty with the United States, their motto being "We prefer a grave in Colombia to a prison in the United States."[

Escobar's campaign of terror made him public enemy number one, forcing him to manage his drug empire while on the run, while turning the streets of Medellin and Bogota into a warzone. Escobar formed an army of poor, teenage sicarios from the ghettos of Medellin and ordered the execution of hundreds of police, paying a $100 bounty for the murder of any member of the CNP. The devastation was total, beyond the wanton execution of cops, Escobar had green-lighted thousands of car bombings, the assassinations of presidential candidate Luis Carlos Galán Sarmiento, and Columbia's attorney general, Carlos Mauro Hoyos Jiménez, along with the downing of a jet airliner, Avianca flight 203, placing a bomb on-board that killed all 103 passengers.

The hunt for Escobar went on for years, encompassing 15,000 raids and thousands of innocent lives, yet he continued to avoid capture, relenting to the public outcry to end the violence, the politicians that ran Columbian government signed off on a deal that saw the drug lord keep his billions and surrender to a prison specially built by Escobar himself, a thinly veiled palace nicknamed "La Catedral," where he, his lieutenants and sicarios were to spend a mere 5 years in the most luxurious jail ever constructed. 

Although the CNP, DEA, and DIJN were devastated by the deal, feeling their efforts to stem the bloodshed and bring down Escobar had been wasted, and the murder of so many left unavenged, they would eventually get a second opportunity to destroy the Medillian cartel and decapitate the head of the snake when Escobar and his people made a daring break from his gilded cage after the Columbian National Army raided the prison in an attempt to transfer Escobar to a maximum security facility after embarrassing the ruling politicians by continuing to run his narcotics empire in brutal fashion, including ordering the deaths of his rivals and personally murdering his own people, from the confines of La Catedral ... and the manhunt was back on.

Up to 50% of U.S. military support, security, and services in Iraq were carried out by contractors, and up to 75% of military-related personnel in Afghanistan have been sourced from the private sector, a reality that would have been unthinkable just two decades earlier. 

The global war on terror, the increasing economic, political, and cultural instability of much of the world's nations, and the exponential quickening of the globe's descent into durable disorder demands that we plan, purchase, and prepare to manage conflicts and win wars with a completely new set of rules, a fresh strategy that recognizes that the wars of the last few hundred years, especially the world wars of the 20th century, are a relic of the past, and that the stealth wars of tomorrow will be ignited and exacerbated by non-state actors, and exploited by the likes of Russia and China with all sides leveraging private armies, contract warfare, and modern-day mercenaries.

World Champion Triathlete Joanna Zeiger:  Mental Toughness, Confidence & the Perfectionism Dilemma

Jeremiah JP Dinnell is a former Navy SEAL Sniper who spent nearly a decade in the SEAL Teams with three combat deployments. J.P. served as Point Man, Machine Gunner, and Lead Sniper for SEAL Team 3, Task Unit Bruiser, Delta Platoon where he worked closely with Task Unit Commander Jocko Wilink, and Leif Babin during the Battle of Ramadi, and was the driving force on many of the combat operations Jocko and Babin wrote about in Extreme Ownership, their groundbreaking book on leadership. As Michael grew into a young man, he became a warrior looking for a war, fueled by violence and an intense need to test his mettle, brawling in bars and on a path to prison, he would eventually rise from being broke and jobless to reenlisting in the military at age 39, two decades after an injury ended his career with the Marines.

We delve deep into Michael's book "We Fight Monsters" a journey into his musings on what it means to be a man and face death, fear, love, war, as well as how to be free, how to dare, and how to be reborn, as seen through the eyes of his journey of self-discovery. Along the way we discuss Michael's views on why cultural elites and "intellectual doubters" seem to be bent on destroying what it means to be a man, fomenting a self-hating, weak society - why men aspire to be Gods and are often plagued by anger, and how we can use the "seeds of pain" born of the trauma wrought by violence, grief and hate to build a positive and rewarding life.
