Our guest this month is Anatol Kreitzer, assistant professor of physiology and neurology at UCSF and a scientist at the UCSF-affiliated Gladstone Institutes. Dr. Kreitzer has made pioneering discoveries in the study of the neural circuits that control movement. His lab is interested in the function of the basal ganglia, a structure deep in the brain that controls movement, motivation, and action selection. Dysfunction of the basal ganglia can lead to movement disorders such as Parkinson’s disease and Huntington’s disease where patients have difficulty either initiating or controlling movements.

To understand how the basal ganglia works, the Kreitzer lab records electrical activity from neurons within the basal ganglia and determines how it relates to movement in behaving mice. They can also control this activity using an emerging technique known as optogenetics. By delivering genes coding for light-sensitive proteins into specific neurons, scientists in the lab can manipulate the electrical activity of certain neurons to see how movement is affected. This technique is being used to study the cells in the basal ganglia that guide our actions based on previous experience. Dr. Kreitzer’s work has provided significant insights into how the basal ganglia works and may eventually lead to potential cures for movement disorders.

The human brain is the most complex structure in the body. It consists of about 100 billion neurons that make around 100 trillion synapses. These connections are constantly changing and the brain must maintain a stable level of electrical activity as it changes. If this balance is disrupted, conditions like epilepsy and schizophrenia can arise. How neurons achieve this feat is still a scientific mystery.

Our guest this month is Dr. Graeme Davis, professor in the department of biophysics at UC San Francisco. Dr. Davis hopes to solve the question of how the brain remains stable as it changes. His lab uses fly genetics to determine important genes involved in maintaining stable neural activity. Listen as Dr. Davis describes how one of these genes, dysbindin, is involved in stabilizing neural function and may have important implications in schizophrenia.

The formation of this extremely complex circuit requires that each neuron find its appropriate target. Dr. David Van Vactor and his lab at Harvard University study the cellular machinery that help motor neurons navigate and find their correct partners, muscles, during development. They are also investigating how the neuromuscular junction is formed and maintained once the neuron reaches its destined target. Diplomatic Immunity

Our immune system is pretty good at hunting down most viruses. But there are a handful of viruses out there that can hide from our immune system for years. The jury is still out on what effect these dormant viruses have on our health. Surprisingly, it might be the case that some of these dormant viruses, like herpes, may actually have some positive benefits. For this piece, producer Meryl Horn talks with professor J.J. Miranda of the Gladstone Institute at UCSF, who explains his innovative approach in investigating this topic.


A Can of Worms

In the late 20th and early 21st centuries, developed countries became increasingly vulnerable to rampant immune system dysfunction, with ballooning rates of allergic and autoimmune disease. Why did this happen? A popular theory is that our hyper-clean environments, and the resulting lack of regular challenge to our immune systems - such as chronic parasitic infections - are causing our immune systems to misbehave. In this episode, producer Sam Ancona Esselmann sits down with Moises Velasquez Manoff,  author of An Epidemic of Absence, to explore this dramatic rise in autoimmune and allergic diseases and to discuss the caveats of controversial therapies. 


T-Cells, 2.0
 
One of the reasons cancer is often so difficult to treat is because cancer cells are winning an “arms race” against our natural defenses. But what if we could give our immune system a tactical advantage? In T-cell immune therapy, T-cells are removed from cancer patients and modified so that they can hunt down specific cancer markers that they were previously unable to recognize. The T-cells, which can then both recognize and kill the cancer cells, are reintroduced into the patient. In this episode, our producer Tyler Ross sits down with scientist Levi Rupp, a member of Wendell Lim's lab at UCSF, who is hacking into our immune cells to fight cancer.

Young Scientist Spotlight 4: Jhia Jackson

A brain is very computationally efficient – you can look at a group of objects and your brain will instantly calculate the average features (size, orientation, etc). But how fast is this process – can it even be done with images that flash by so quickly you aren’t sure if you even saw them? To learn more, we interviewed Maria Servetnik, a Master’s student at the University of Leuven in Belgium who is currently testing this hypothesis. In our conversation, we also discussed the importance of mentorship, her journey from journalism to science, the challenges of communicating science to people who don’t trust the scientific process, and how to stay sane amidst the combined struggles of grad school and 2020.

Science is one of the most powerful influences in our lives. The medicines we use, the foods we eat, the technologies we have – they are all products of the scientific process. But many people are disconnected from that process. They see science as something foreign, done in secret labs by strange people. And that’s wrong! Science is an open discussion, and scientists are regular folk who happen to be curious about how things work. But how can scientists show that to the public?

CTOR is a podcast designed to bridge the gap between scientists and the general public. We give scientists a platform to talk about their work, their motivations for being scientists, and what gets them out of bed in the morning. We try to find researchers who are passionate about their work, and who are operating at the edge of their field. That way, listeners get to hear about cool discoveries directly from the researchers (sometimes even before the discoveries are made public). We post 1-2 news episodes each month, along with information about the scientists we interviewed. And because we let the scientists speak directly to the public, listeners get more than sound bites and pretty pictures; they get the whole story.
