The evening light slowly sinks with the stillness of the lake, and the night is getting darker.

The air becomes colder, the gusts of wind subside, and the body becomes lighter. The feeling of weightlessness lifts up the souls in the bodies of sleeping people. They are like transparent moths chasing the bright crescent moon in the distance, flying faster and faster.

They said they dreamed last night that they were back in a place that felt very familiar. It was pure, bright, and a little warm, just like the reverse-colored night sky.

The naming of yeast strains is less formalized, but generally includes an alphanu-
meric combination that is unique to the laboratory of origin, followed by the genotype.
For example, BY4742 (MAT a his3D1 leu2 D0 lys2 D0 ura3 D0) represents the haploid
strain BY4742, which is of the a mating type and auxotrophic for histidine, leucine,
lysine, and uracil biosynthetic pathways. When a gene is expressed from an intro-
duced plasmid rather than its natural chromosomal location, this is noted by
inclusion of the plasmid name with associated genes. Multicopy plasmids contain
a DNA replication origin from the endogenous yeast 2 micron circle plasmid (often
represented as YEp followed by a specific number, YEplac181 [1]) or from a
chromosomal autonomous replication sequence (often represented as YRp). The
addition of a yeast centromere (often represented as YCp) greatly enhances the
plasmids mitotic and meiotic stabilities and reduces the copy number to approxi-
mately one copy per haploid cell.
Most laboratory yeast cultures originate from strain S288C and a handful of other
progenitor isolates [2] whose derivatives have been used to complete the yeast genome
sequence and to create extensive library collections of knockout mutants [3], hem-
agglutinin-tagged [4], TAP-tagged [5], and GFP-tagged [6] strains, bar-coded
strains [7,8], and sets of strains bearing single inducible [9], repressible [10], or
hypomorphic [11] gene derivatives. These have proven invaluable tools for high-
throughput studies of gene function and systems biology approaches to model
complex biological networks. Among the commercial vendors of yeast strain collec-
tions are the American Type Tissue Collections (ATCC), InvitrogenÒ, and Open
BiosystemsÒ.
