Hypoxia, a potentially fatal shortage of oxygen in tissues, is a complicating factor in a variety of serious illnesses including cancer, cardiovascular disease, and respiratory conditions. Diffusion Pharmaceuticals lead experimental candidate, TSC, enhances the ability of the body to deliver oxygen to where it is needed most. We spoke to Bob Cobuzzi, CEO of Diffusion Pharmaceuticals, about hypoxia, the company’s lead therapeutic candidate to address it in a broad range of conditions, and its clinical development strategy for the drug.

Delivering Oxygen to Tissue in Need

A song composed from the sonification of chemical data gleaned from the ear of a Baltic cod that lived during the stone age, 4,500-5,000 years ago.  An "earstone", a calcium carbonate part of the cod's ear, was unearthed at an archeological site on Gotland Island in the Baltic sea and was chemically analyzed by scientists. The chemical composition of the earstone reveals details about the fish's life and the quality of the water where it swam.  

The chemicals suggest that this fish grew well and lived in clean water. Fishes are described as "happy" if the data from the chemical trace elements present in their earstones indicate that they lived in good quality water, and "unhappy" if the data indicates they encountered low-oxygen conditions during their lifetimes. Tate IV to answer these questions; give a behind-the-scenes look at an oceanographer's life both on deck and below; and describe her journey to being elected to the most prestigious scientific society in the U.S., the National Academy of Sciences. For more than three decades, Dr. Rabalais’ determination to protect marine life in the gulf has resulted in several appearances before Congress and 35 research cruises. Dr. Rabalais is a professor and the Shell Endowed Chair in Oceanography and Wetland Studies in the Department of Oceanography & Coastal Sciences at Louisiana State University.

Hypoxemia occurs when levels of oxygen in the blood are lower than normal. If blood oxygen levels are too low, your body may not work properly. Blood carries oxygen to the cells throughout your body to keep them healthy. When hypoxemia causes low oxygen levels in your body’s tissues, it is called hypoxia. Common hypoxia symptoms include confusion, cough, fast heart rate, rapid breathing, shortness of breath, sweating, and wheezing.

You need to go to the hospital to get treatment for hypoxia and to keep a check on your oxygen level. The most important thing is to get more oxygen into your body. You may receive it through a small plug in your nose or through a mask that covers your nose and mouth. For many people, this is enough to bring your oxygen level up to normal. Frequencies of 30, 460, 600, 850, 2500, 5250, 17500, 93500, 224940, 425610 Hz can alleviate hypoxia symptoms.

A small regulatory protein can be found not only in all humans, animals, plants and fungi, but in each and every eukaryotic cell. Following the discovery of these ubiquitous proteins in 1974, researchers aptly named them ubiquitin. Enzymes and/or ligases (enzymes that catalyze the formation of chemical bonds), allow ubiquitins to bind to substrate proteins. This process, known as ubiquitination, can initiate or prevent protein interactions, denote them for degradation and alter their activity and cellular location.

Aberrant ubiquitination may play an important role in tumorigenesis. At the core of solid tumors, hypoxia-inducible factor 1α (HIF-1α) is induced (by hypoxia) and allows cancer cells to adapt to the lack of oxygen. The accumulation of HIF-1α can also transcribe a number of genes well-known to be involved in cancer. Therefore, HIF-1α is an attractive, albeit difficult-to-pinpoint, therapeutic target in cancer. Researchers—from Brown University and ​​Lifespan Cancer Institute—previously observed the stabilization of HIF-1α by the protein coding gene cyclin-dependent kinases CDK1. They further proposed that CDK4 may also be an HIF-1α stabilizer. However, the mechanism of HIF-1α’s regulation by CDK4 has yet to be fully elucidated. 

“Till now, development of therapies targeting HIF-1α remains hindered. Therefore, it is imperative to explore the mechanism of HIF-1α regulation in cancer cells and investigate new possibilities to therapeutically target HIF-1 signaling.”

In 2021, the same team conducted a new study further investigating the molecular mechanisms of HIF-1α destabilization by CDK1 or CDK4/6 inhibitors in colorectal cancer. Their priority research paper was published as the cover of Oncotarget’s Volume 12, Issue 20, and entitled, “Identification of Smurf2 as a HIF-1α degrading E3 ubiquitin ligase.”

New Study: Protein Analysis Reveals Novel Role of Enzyme In Cancer

The CLASS Acts podcast is an initiative of UCD Conway Institute, a research institute based in University College Dublin.

In episode 4, Dr. Eoin Cummins talks with Assistant Professor Sergio Rey, Ad Astra Fellow in the UCD School of Medicine who is a Conway Fellow. 

Sergio describes how he trained as a clinician scientist in Chile before persistence and hard work led him to a Nobel prize winning laboratory in John Hopkins University. Sergio’s interest in hypoxia and cancer subsequently led him to Dublin (via Canada) where he is excited about the next chapter in his research career as an Ad. Astra fellow.

He was inspired to create the CLASS Acts podcast to share the many and varied backgrounds and journeys taken by Conway researchers on their routes to scientific success.

Hailing from London, Upon You's 2nd time signing for Hypoxia LDN steps up with his anticipated debut three-tracker, "Into The River". Operating the grey zone between no-nonsense jacking house motifs and subaquatic atmospheres wrapped in thick, reverb-drenched membranes, the British producer steers us amidst a sonic universe both hi-tech and organically suave, meditative and undeniably functional.

So works the opener "Gream", a proper hypnotic slab of clinical programming and bass-heavy charge slowly moving onto broader, further thunderous minimalism as it finishes us off to a feast of raucous dub echoes and heavily processed machine bombast. Then comes the hi-NRG shuffle of "The Not Fine Art Of Repetition", Hypoxia's subtle touch making wonders in carving out that infectiously jagged groove out seemingly prototypical tech-house textures and, again, that ominous bass relief, while vocals beckon us onto unmistakably UK garage territories.

Last but far from least, title-track "Into The River" injects a heathy dose of jazz and boogie intricacies into the equation, trading the relative austerity of the previous for a funkier, disco-infused approach. The result is a soulful chugger that lacks neither peak time party oomph nor that jiggy swagger and wholeheartedness, making it both a perfect number for transitional sequences both on and off the dance floor, when the mind and body seem to float in a distinct haze and recombine under the friendly yoke of the almighty groove.

Born in 2001, in the sunshine state Florida, Hypoxia started making music at the age of 11. Inspired by  EDM artists such as Darude, Deadmau5, and Hardwell, Hypoxia started downloading synthesizers. Experimenting with sound design and producing, a burning passion was lit.

My objective is to share music that has been influential to me through the creative act of conceptual, audio-journaling. 

A deep interest in metamorphosis has evolved into my podcasts. This brings together various styles of sound, while breaking down the walls of genre into pure visionary realms.

It is the year 2007.

Electronic music is more popular than ever, especially TechHouse is on a thriving uprise and Berlin already became the buzzing nightlife capital of the world it is known as today.

Amidst all these creative and musical vibes a new label is born -  Upon.You Records. Founded in February 2007 by DJ & producer Marco Resmann, with a first release under his moniker Phage on Martin Landsky's Intim Recordings as well as great success as one half of the infamous project Luna City Express under his belt, and electronic music veteran Hawks Grunert, formerly involved in running Timing Recordings with Tanith as well as working for legendary „Frontpage“ magazine, Upon.You Records was already widely recognized for its first release, Phage's „Vut & Vat EP“, and started to drive on the fast lane immediately.

Originally intended to be a platform for music from an inner circle of friends as well as for aspiring young talent it didn't take long until the label branched out in 2008 with first international signings,  various label tours all over the place and a first label compilation.

In 2009, after receiving massive support from nightlife heavyweights like Steve Bug, Brothers Vibe, Someone Else and many more, a cover story in German electronic music magazine GROOVE and with a catalogue of roughly twenty 12“ releases Upon.You launched two subdivisions - Upon.You Digital and the momentary Upon.You Sweet & Sour Series – and established a booking agency as well as regular label events at two of Berlin's most praised venues, Watergate Club and the infamous Panoramabar.

Furthermore Upon.You Records teamed up with Workaholics Berlin for the launch of a merchandising line in 2009, introducing the labels unique visual image to the world of street fashion.

With the launch of the annual label retro- / futurespective „Upon.You Diary“ and an expanded core artist family now including Marco Resmann, Martin Landsky, ONNO, Ruede Hagelstein, Gunnar Stiller, Magit Cacoon and The Cheapers alongside many others, 2010 finally paved the way for success and fruitful growth to come in consecutive years in which Upon.You became one of the leading labels in all things TechHouse and related.

And it was even the first issue „Upon.You Diary“ that awed the electronic world with Marco Resmann's widely praised tune „This Song“, immediately ruling every dancefloor after it was released and drawing even more attenttion towards the Berlin-based label which continued to expand its growing international network whilst signing releases by Quenum, Kink or the UK-based longtime ambassador Steve Lawler.

On a jam-packed schedule of about twenty releases per year lately, including Ruede Hagelstein's Beatport smash „Minus“, with label artists touring the globe frequently and armed with the brand new, 2014-launched UYX subdivision focusing on raw TechHouse and classic stamped whitelabel aesthetics, Upon.You has grown into a major force on the international electronic music circuit.

Always on the lookout for new talent and keen to explore new territories, the future looks brighter than ever for Upon.You after entering the longplay circuit with the widely acknowledged album debut of the former Tel Aviv- and now Berlin-based DJ / producer Magit Cacoon, another successful instalment of the „Upon.You Diary“-series and finally entering three digit realms with the release of Marco Resmann's extrordinary gold vinyl 12“  „100“ - the onehundredth Upon.You single release in the label history which was released in late 2015.

And things surely are about to be busier than ever for all members of the Upon.You camp over the course of 2016, celebrating the labels 8th anniversary with a dedicated series of label events all over Europe and special showcases at Sonar / Barcelona as well as ADE and, of course, a string of high quality releases for maximum dancefloor pleasure throughout this season and beyond.
