デングウイルス感染症は蚊を媒介して年間1億人が感染し、二回目の感染で重症化しやすいが、有効な治療薬が未だ開発されていないため、ワクチン接種による感染防止が一番有効な対策と考えられる。そこで本研究では、組換え蛋白質ワクチンの候補であるDEN4 ED3（4型デングウイルス由来エンベロープ蛋白質第３ドメイン）に注目し、加熱・攪拌・凍結融解などバイオ医薬品の製造プロセスで起こりうるストレスを加えたとき、蛋白質凝集による品質低下が誘導されるか検証する。
　先行研究ではDEN4 ED3の熱凝集の前駆体である可逆的なオリゴマー（RO)をモデル蛋白質とし、疎水性アミノ酸（３８０番目のバリン）を一残基置換するだけで、酸性ｐH・高温でのRO形成の阻害に成功した。そこで本研究では、 DSC（示差走査熱量計）やDLS（動的光散乱）などの物性測定を実施し、生理的条件下での加熱以外のストレスでも一残基置換による蛋白質凝集の抑制効果が得られるか確認する。
