BUFFALO, NY- December 15, 2023 – A new #researchpaper was #published on the #cover of Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 15, Issue 23, entitled, “Uncovering a unique pathogenic mechanism of SARS-CoV-2 omicron variant: selective induction of cellular senescence.”

SARS-CoV-2 variants are constantly emerging with a variety of changes in the conformation of the spike protein, resulting in alterations of virus entry mechanisms. Solely omicron variants use the endosomal clathrin-mediated entry. 

In this new study, researchers Franziska Hornung, Nilay Köse-Vogel, Claude Jourdan Le Saux, Antje Häder, Lea Herrmann, Luise Schulz, Lukáš Radosa, Thurid Lauf, Tim Sandhaus, Patrick Samson, Torsten Doenst, Daniel Wittschieber, Gita Mall, Bettina Löffler, and Stefanie Deinhardt-Emmer from Jena University, Leibniz Centre for Photonics in Infection Research (LPI), University of California San Francisco, Klinik für Herz- und Thoraxchirurgie, and University Hospital Bonn investigated the influence of defined altered spike formations to study their impact on premature cellular senescence.

“In our study, in vitro infections of SARS-CoV-2 variants delta (B.1.617.2) and omicron (B.1.1.529) were analyzed by using human primary small alveolar epithelial cells and human ex vivo lung slices. We confirmed cellular senescence in human lungs of COVID-19 patients. Hence, global gene expression patterns of infected human primary alveolar epithelial cells were identified via mRNA sequencing.”

Solely omicron variants of SARS-CoV-2 influenced the expression of cell cycle genes, highlighted by an increased p21 expression in human primary lung cells and human ex vivo lungs. Additionally, an upregulated senescence-associated secretory phenotype (SASP) was detected. Transcriptomic data indicate an increased gene expression of p16, and p38 in omicron-infected lung cells. Significant changes due to different SARS-CoV-2 infections in human primary alveolar epithelial cells with an overall impact on premature aging could be identified. A substantially different cellular response with an upregulation of cell cycle, inflammation- and integrin-associated pathways in omicron infected cells indicates premature cellular senescence.

“This difference may be attributed to the distinct endocytic cell entry and intracellular pathways of the omicron variant when compared to the delta variant. The induction of cellular senescence in lung tissue following acute SARS-CoV-2 infection could potentially contribute to the reported cytokine storm and the development of long-COVID.”

La Comisión independiente de Investigación sobre la Pandemia de Covid-19 en México declaró que se calcula que ocurrieron 300 mil muertes adicionales durante la pandemia, las cuales se atribuyen a errores de gestión gubernamental. Entre estos errores se incluyen la deficiente explicación sobre la transmisión del virus, la falta de promoción del uso de cubrebocas, la insuficiente inversión en salud pública y la escasez de apoyos para la ciudadanía, entre otros factores.

BUFFALO, NY- May 6, 2024 – A new research paper was published in Oncotarget's Volume 15 on May 3, 2024, entitled, “Transfected SARS-CoV-2 spike DNA for mammalian cell expression inhibits p53 activation of p21(WAF1), TRAIL Death Receptor DR5 and MDM2 proteins in cancer cells and increases cancer cell viability after chemotherapy exposure.”

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and COVID-19 infection has led to worsened outcomes for patients with cancer. SARS-CoV-2 spike protein mediates host cell infection and cell-cell fusion that causes stabilization of tumor suppressor p53 protein. In-silico analysis previously suggested that SARS-CoV-2 spike interacts with p53 directly but this putative interaction has not been demonstrated in cells. In this new study, researchers Shengliang Zhang and Wafik S. El-Deiry from Brown University and Lifespan Health System examined the interaction between SARS-CoV-2 spike, p53 and MDM2 (E3 ligase, which mediates p53 degradation) in cancer cells using an immunoprecipitation assay. 

“We observed that SARS-CoV-2 spike protein interrupts p53-MDM2 protein interaction but did not detect SARS-CoV-2 spike bound with p53 protein in the cancer cells.”

The researchers further observed that SARS-CoV-2 spike suppresses p53 transcriptional activity in cancer cells including after nutlin exposure of wild-type p53-, spike-expressing tumor cells and inhibits chemotherapy-induced p53 gene activation of p21(WAF1), TRAIL Death Receptor DR5 and MDM2. The suppressive effect of SARS-CoV-2 spike on p53-dependent gene activation provides a potential molecular mechanism by which SARS-CoV-2 infection may impact tumorigenesis, tumor progression and chemotherapy sensitivity. In fact, cisplatin-treated tumor cells expressing spike were found to have increased cell viability as compared to control cells. 

Further observations on γ-H2AX expression in spike-expressing cells treated with cisplatin may indicate altered DNA damage sensing in the DNA damage response pathway. The preliminary observations reported here warrant further studies to unravel the impact of SARS-CoV-2 and its various encoded proteins including spike on pathways of tumorigenesis and response to cancer therapeutics. More efforts should be directed at studying the effects of the SARS-CoV-2 spike and other viral proteins on host DNA damage sensing, response and repair mechanisms. 

“A goal would be to understand the structural basis for maximal anti-viral immunity while minimizing suppression of host defenses including the p53 DNA damage response and tumor suppression pathway. Such directions are relevant and important including not only in the context of viral infection and mRNA vaccines in general but also for patients with cancer who may be receiving cytotoxic or other cancer treatments.”

Transfected SARS-CoV-2 Spike DNA Suppresses Cancer Cell Response to Chemotherapy

The profound impact of the COVID-19 pandemic, caused by the SARS-CoV-2 virus, has been felt across various domains, including the realm of cancer research and treatment. As scientists delve deeper into the intricate mechanisms of this viral pathogen, intriguing revelations have emerged regarding its potential influence on cellular processes pivotal to cancer development and progression. 

In a new study, researchers Wafik S. El-Deiry and Shengliang Zhang from Brown University and Lifespan Health System shed light on the intricate interplay between the SARS-CoV-2 spike protein and the tumor suppressor p53, a key guardian of genomic integrity. On May 3, 2024, they published their new research paper in Oncotarget’s Volume 15, entitled, “Transfected SARS-CoV-2 spike DNA for mammalian cell expression inhibits p53 activation of p21(WAF1), TRAIL Death Receptor DR5 and MDM2 proteins in cancer cells and increases cancer cell viability after chemotherapy exposure.”

In this study, researchers El-Deiry and Zhang uncovered a hitherto unknown facet of the SARS-CoV-2 spike protein’s impact on cancer cells. Their findings, meticulously detailed in their research paper, have ignited a newfound curiosity within the scientific community.

SARS-CoV-2 Spike Protein Disrupts p53 Tumor Suppressor Pathway

BUFFALO, NY- May 21, 2024 – A new research #paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 9, entitled, “Germicidal lamps using UV-C radiation may pose health safety issues: a biomolecular analysis of their effects on apoptosis and senescence.”

The battle against the COVID-19 pandemic has spurred a heightened state of vigilance in global healthcare, leading to the proliferation of diverse sanitization methods. Among these approaches, germicidal lamps utilizing ultraviolet (UV) rays, particularly UV-C (wavelength ranging from 280 to 100 nm), have gained prominence for domestic use. 

These light-emitting diode (LED) lamps are designed to sanitize the air, objects, and surfaces. However, the prevailing concern is that these UV lamps are often introduced into the market without adequate accompanying information to ensure their safe utilization. Importantly, exposure to absorbed UV light can potentially trigger adverse biological responses, encompassing cell death and senescence.

In this new study, researchers Nicola Alessio, Alessia Ambrosino, Andrea Boggi, Domenico Aprile, Iole Pinto, Giovanni Galano, Umberto Galderisi, and Giovanni Di Bernardo from the University of Campania Luigi Vanvitelli, Regional Public Health Laboratory in Siena, Italy, ASL Napoli 1 Centro P.S.I. Napoli Est-Barra, and Temple University performed a series of investigations aimed at comprehending the biological repercussions of UV-C radiation exposure from readily available domestic lamps. 

“Our focus centered on epithelial retinal cells, keratinocytes, and fibroblasts, components of the skin and ocular targets frequently exposed to UV irradiation.”

Their findings underscore the potential harm associated with even brief exposure to UV, leading to irreversible and detrimental alterations in both skin cells and retinal cells of the eye. Notably, epithelial retinal cells exhibited heightened sensitivity, marked by substantial apoptosis. In contrast, keratinocytes demonstrated resilience to apoptosis even at elevated UV doses, though they were prone to senescence. Meanwhile, fibroblasts displayed a gradual amplification of both senescence and apoptosis as radiation doses escalated.

“In summary, despite the potential benefits offered by UV-C in deactivating pathogens like SARS-CoV-2, it remains evident that the concurrent risks posed by UV-C to human health cannot be ignored.”

El Dr. Antonio Lazcano nació en Tijuana, Baja California, el 10 de abril de 1950. Estudió la licenciatura de Biología en la Facultad de Ciencias de la UNAM, donde posteriormente obtuvo el grado de doctor en Ciencias. Desde abril de 2002 se desempeña como Profesor Titular “C” de Tiempo Completo en la misma Facultad y donde fundó el curso optativo “Origen de la vida” y el laboratorio con el mismo nombre. También ha trabajado como profesor residente y científico visitante en Francia (Universidad de Orsay París-Sud, Instituto Pasteur de París), España (universidades de Alicante, Autónoma de Madrid, Valencia), Cuba (Universidad de La Habana), Italia (Universidad de Roma “La Sapienza”), Suiza (ETH Zentrum de Zurich), Rusia (Instituto A. N. Bakh de Bioquímica de Moscú), y en los Estados Unidos (Universidad de California en San Diego y Universidad de Houston).

Se ha especializado en el estudio del origen y la evolución temprana de la vida por medio del análisis de meteoritos y de simulaciones experimentales de la Tierra primitiva, así como mediante del análisis de secuencias de genes muy antiguos. Durante los últimos tiempos se ha dedicado, entre otras líneas de investigación, a caracterizar las propiedades de los diferentes tipos de RNA polimerasas celulares y virales para identificar algunos componentes más antiguos, inclusive, que el DNA. Dirige, en forma honoraria, el Centro “Lynn Margulis” de Biología Evolutiva en las Islas Galápagos, Ecuador.

Es autor de más de 150 trabajos de investigación publicados en revistas con arbitraje internacional, alrededor de 70 artículos de divulgación científica, y es autor o coautor de 16 libros especializados sobre problemas evolutivos. Algunas de sus obras más conocidas son La bacteria prodigiosa (1988), Alexander I. Oparin: La chispa de la vida (2008), y El origen de la vida: evolución química y evolución biológica (1983).

Entre los reconocimientos que ha obtenido el Dr. Lazcano Araujo están el de Investigador Nacional Nivel III por parte del Conacyt, y la Distinción “Catedrático Universitario” por parte de la UNAM. En 1991 recibió, junto con el Dr. José Sarukhán, la primera Medalla a la Investigación Biológica “Alfonso L. Herrera”, otorgada por la Escuela de Biología de la Universidad Autónoma de Puebla; durante el periodo 1997-1998 formó parte del Comité Científico organizado por la NASA para supervisar la creación del Astrobiology Institute.

Otras distinciones incluyen su elección como Vicepresidente (1997-1999), primero, y luego Presidente (1999-2001) de la Gordon Conference of the Origins of Life, además de haber sido dos veces Presidente de la International Society for the Study of the Origins of Life, convirtiéndose en el primer científico latinoamericano en llegar a ese puesto, al que fue reelegido. En 2006 recibió la Medalla de la Gran Cruz de Fundador de la Universidad de San Francisco de Quito; en 2007 la UNAM le otorgó el Premio Universidad Nacional en Investigación en Ciencias Naturales, y al año siguiente la Universidad de Milán le concedió el Doctorado Honoris Causa, seguido unos meses mas tarde por la Primer Medalla “Francesco Redi”, que le otorgó la Sociedad Italiana de Astrobiología. En 2009 recibió de la Universidad Veracruzana la Medalla al Mérito Universitario, y en 2013 la Tercera Cumbre de la Evolución le otorgó en las Islas Galápagos el “Charles Darwin Distinguished Scientist Award” junto a la Dra. Ada Yonath, Premio Nobel de Química (2009). En mayo de 2015 la Universidad de Valencia, España, le confirió el grado de Doctor Honoris Causa.

El Dr. Antonio Lazcano Araujo ingresó a El Colegio Nacional el 6 de octubre de 2014. Su discurso de ingresó se tituló “La aparición de la vida: consiliencias y discordancias”, y fue contestado por el doctor José Sarukhán.
